In order to base-pair with a strand of DNA or RNA, oligonucleotides are created. Oligonucleotides are most frequently used as polymerase chain reaction primers. The 5' end of the sequence intended for amplification serves as the complement to at least part of the sequence used in the construction of primers. The primers anneal to the complementary sequences on the DNA templates during the annealing process.
